When chelating agents are present in the sample, the medium minimizes nickel ion leaching while maximizing protein recovery. It is suitable for purifying proteins expressed in eukaryotic cells like insect-baculovirus, 293 cells, and CHO cells, as well as in bacteria such as E. coli. The medium allows direct loading of samples without pre-treatment, preventing nickel leaching. Ni Agarose FF / Ni Agarose HP affinity chromatography media are designed by chelating divalent nickel ions (Ni(2+)) onto an agarose gel matrix with aminotriacetic acid as the ligand. These media offer several advantages, including high adsorption capacity, good selectivity, ease of regeneration, and low cost.
