Description
CCN Bio SUMO Protease is a recombinant protease derived from the Saccharomyces cerevisiae ubiquitin-like-specific protease 1 (ULP1). The enzyme specifically recognizes the three-dimensional structure of the SUMO (Small Ubiquitin-like Modifier) fusion tag and cleaves precisely at the junction between the SUMO tag and the target protein.
Unlike sequence-specific proteases, SUMO Protease recognizes the folded SUMO domain rather than a short amino acid sequence. This unique mechanism provides exceptionally high specificity and enables cleavage without leaving additional amino acid residues on the N-terminus of the target protein.
CCN Bio SUMO Protease exhibits robust activity across a broad range of reaction conditions, including pH 7.0–9.0 and temperatures from 4°C to 37°C. The enzyme is compatible with many commonly used purification buffers and tolerates moderate concentrations of additives such as imidazole, EDTA, urea, and detergents.
The SUMO fusion expression system is widely recognized as one of the most effective approaches for improving soluble expression of recombinant proteins in bacterial hosts. Compared with traditional fusion partners such as ubiquitin, GST, MBP, and Trx, SUMO often provides superior protein solubility, stability, and expression yields.
CCN Bio SUMO Protease contains an N-terminal His-tag, allowing convenient removal of the protease following digestion using Ni-NTA affinity resins or magnetic beads.
This product is supplied without a digestion buffer, enabling users to optimize reaction conditions for individual target proteins.
